DTT is a popular reducing agent for many protein research applications, including preparing proteins for reducing polyacrylamide electrophoresis (SDS-PAGE) and reducing disulfide bonds for labeling sulfhydryl groups in conjugation reactions. At low concentrations, DTT is used to stabilize proteins and enzymes with free sulfhydryl groups to keep monothiols in a reduced state. DTT reduction is also used to prepare proteins for mass spectrometry.

DTT is not stable in solution and should be prepared fresh for each use. The No-Weigh format consists of a 48-microtube tray containing dry, stable, single-use aliquots of the DTT chemical to eliminate the need to weigh small amounts of reagent and prevent waste of unused DTT. Just puncture the seal of one microtube, add 100 μL of water or buffer, and mix to dissolve. In just a few seconds you will have a solution of 500 mM DTT, ready for a variety of laboratory methods. Dilute as necessary or add the appropriate volume of the 500 mM stock solution to achieve the amount of DTT reducing agent required for your specific application.
