Description
Acryloyl-X, SE (6-((acryloyl)amino)hexanoic acid, succinimidyl ester) is an amine-reactive molecule that enables crosslinking of acrylamide to proteins and other biomolecules containing amines. The succinimidyl ester of acryloyl-X covalently reacts with amines of proteins, amine-modified nucleic acids, and other biomolecules to yield acrylamides that can be copolymerized into polyacrylamide matrices or on surfaces, such as in microarrays and in biosensors. For instance, acryloyl-X, SE is used to prepare streptavidin acrylamide (Cat. No. S21379), which can copolymerize with acrylamide on a polymeric surface to create a uniform monolayer of immobilized streptavidin for binding to biotinylated ligands such as hybridization probes, enzymes, antibodies, and drugs.
Acryloyl-X, SE is a critical crosslinking reagent used in the super-resolution imaging techniques expansion microscopy (ExM) and protein-retention expansion microscopy (proExM) to add acrylamides to the amines of proteins of a preserved cell or tissue sample. This crosslinking with acrylamide allows the proteins to be anchored to a polymer hydrogel and polymerized. The polymer hydrogel is then swelled, allowing the cell or tissue sample to be physically expanded to improve spatial resolution in imaging. Expansion microscopy is a versatile super-resolution imaging technique that can be performed on many specimens, including 3D imaging of thick samples. This imaging technique also uses conventional microscopes, enabling easier and faster visualization of nanoscale biological structures without advanced super-resolution microscopes and equipment.
