Description

OMA1-03061 detects protein kinase B (PKB) phosphorylated at Ser473 from human and mouse samples. OMA1-03061 detects PKB 1,2,and 3. OMA1-03061 has been successfully used in Western blot, immunofluorescence, and immunocytochemistry procedures. By Western blot, this antibody detects an ∽55 kDa protein representing phosphorylated PKB (Ser473) from NIH-3T3 cells treated with platelet-derived growth factor (PDGF). The OMA1-03061 immunizing phosphopeptide was derived from a region of human PKB that contains serine 473. There is 100% homology with other species, that have not yet been tested. Antibodies to this protein (and modification) were previously sold as part of a Thermo Scientific Cellomics High Content Screening Kit. This replacement antibody is now recommended for researchers who need an antibody for high content cell based assays. It has been thoroughly tested and validated for cellular immunofluorescence (IF) applications. Further optimization including the selection of the most appropriate fluorescent Dylight conjugated secondary antibody may have to be performed for your high content assay.

AKT1 (PKB alpha) is a serine/threonine kinase that regulates cell survival. The activated enzyme inhibits apoptosis and stimulates cell cycle progression by phosphorylating numerous targets in various cell types, including cancer cells. This protein kinase is activated by insulin, PI3K, IGF1 and various other growth and survival factors. Akt promotes cell survival by inhibiting apoptosis through phosphorylation and inactivation of several targets, including forkhead transcription factors, and caspase-9. The AKT pathway is a major target for cancer drug discovery.
